چکیده

A thin-layer chromatography-gas-liquid chromatography system has made it possible to separate the individual tocopherols for qualitative and quantitative determination in blood plasma and red blood cells. The peak of the individual tocopherols found in plasma and red blood cells were identified by their RF values and comparing their relative retention times with that of 5,7-dimethyltocol which served as an internal reference standard. The detection limits for the tocopherol with the hydrogen flame detector was found to be in the range of 0.03 ,ug. Fat-soluble reducing substances which constitute the major source of interference in the commonly used methods do not interfere with the separation of the tocopherols.

Spectrophotometric method for determination of tocopherol in red blood cells.

A relatively rapid procedure is described for the spectrophotometric determination of total tocopherol in red blood cells (RBC) based on a modification of the original Emmerie-Engel reaction. The critical feature in this method is the presence of a large amount of an added antioxidant, pyrogallol or ascorbic acid, during the saponification and extraction stages and the use of thin-layer chromat...

Determination of valproic acid in human plasma using dispersive liquid-liquid microextraction followed by gas chromatography-flame ionization detection

Objective(s): Dispersive liquid-liquid microextraction coupled with gas chromatography (GC)-flame ionization detector was developed for the determination of valproic acid (VPA) in human plasma. Materials and Methods: Using a syringe, a mixture of suitable extraction solvent (40 µl chloroform) and disperser (1 ml acetone) was quickly added to 10 ml of diluted plasma sample containing VPA (pH, 1....

Human plasma derived drugs separation by fractionation of plasma with polyethylene glycol

Background: There are varieties of purification techniques for separation of human plasma proteins such as salting out, ion exchange chromatography, and ethanol fractionation. There are limitations for each method, for example in salting out method, the salt has to be removed in an additional step. Ion exchange chromatography is difficult for scaling up, and plasma fractionation is a time consu...

Polyunsaturated fatty acids are lower in blood lipids of Usher's type I but not Usher's type II.

PURPOSE Previous studies have shown that persons with retinitis pigmentosa and Usher's syndrome have lower blood levels of long-chain polyunsaturated fatty acids (PUFAs). In this study, the fatty acid composition of phospholipids from plasma and red blood cells (RBCs) was compared in persons with Usher's syndrome type I; Usher's syndrome type II; or no retinal disease (control subjects). METH...

Preparation of Plasminogen by Affinity Chromatography

Background: Plasminogen is one of the compounds derived from human plasma. Activation of plasminogen produces plasmin. Plasmin is able to lyse fibrinogen, fibrin, and some other human plasma proteins. The aim of the present work was to study the separation of human plasminogen by affinity chromatography using gel lysine Sepharose. Materials and Methods: Normal human plasma was used as the...
